تعیین ژنوتایپ گونه های فاسیولا جدا شده از حیوانات اهلی در غرب ایران با استفاده از روش PCR-RFLP

<span style="font-size:12pt"><span style="direction:rtl"><span style="unicode-bidi:embed"><span sans-serif="" style="font-family:Arial,"><span lang="AR-SA" style="font-size:14.0pt"><span b="" lotus="" style="font-family:">فاسیولیازیس که عامل آن انگل فاسیولا هپاتیکا و ژیگانتیکا می­باشد بیماری مشترک بین انسان و حیوانات است. طبق آمارهای معتبر حدود 17 میلیون نفر در دنیا به این انگل آلوده بوده و سالیانه خسارات جبران ناپذیری را به مراکز دام پروری و بهداشتی وارد می­کند. تشخیص رایج این دو گونه بر مبنای خصوصیات مورفولوژیک کرم بالغ و تخم انجام میگیرد. با توجه به تاریخچه طولانی فاسیولازیس در ایران و بروز دو اپیدمی بزرگ آن در استان گیلان (رشت و بندر انزلی) نیاز به یک روش آسان، سریع و قابل اعتماد برای تفکیک گونه­های انگل فاسیولا جهت تفکیک هر چه دقیق تر این دو انگل و تعین ژنوتایپینگ وتوالی آنها جهت بررسی تفاوت­های احتمالی زیر گونه­ای وآنگاه درک بهتر پاتولوژی و اپیدمیولوژی آنها ضروری به نظر میرسد</span></span><span dir="LTR" style="font-size:14.0pt"><span new="" roman="" style="font-family:" times="">.</span></span><span lang="AR-SA" style="font-size:14.0pt"><span style="font-family:"B Lotus""></span></span></span></span></span></span><br>

<span dir="RTL" lang="AR-SA" style="font-size:14.0pt"><span b="" lotus="" style="font-family:">تفکیک گونه­ها و زیر گونه­های انگل به خصوص در کشورهایی که گسترش انگل روی هم افتادگی (</span></span><span style="font-size:14.0pt"><span new="" roman="" style="font-family:" times="">Overlap</span></span><span dir="RTL" lang="AR-SA" style="font-size:14.0pt"><span b="" lotus="" style="font-family:">)دارد مانند ایران، مصر، پاکستان و کشورهای آسیای جنوب شرقی مشکلات فراوانی را ایجاد کرده است و در موارد بسیاری محقق مجبور به حذف نمونه­ای است که با زحمت فراوان تهیه کرده است. تفکیک گونه­های فاسیولا هپاتیکا از فاسیولا ژیگانتیکا و گونه­های حد واسط (</span></span><span style="font-size:14.0pt"><span new="" roman="" style="font-family:" times="">Intermediate</span></span><span dir="RTL" lang="AR-SA" style="font-size:14.0pt"><span b="" lotus="" style="font-family:">) با استفاده از روش­های مولکولی ساده تر و معتبرتر است. تکنیک های مختلف مرفومتریک و ژنوتایپیک در سال­های اخیر برای تمایز گونه و زیر گونه­های فاسیولا کاربرد فراوان داشته است. از طرفی توالی نوکلوتید های تعیین شده در منطقه مزبور و میزبان های قید شده را میتوان با توالی های قید شده در سایر نواحی مقایسه نمود و نهایتا پلی مرفیسم انگل را در مناطق مختلف و میزبان های مختلف بررسی کرد.</span></span>

the current study was to determine the Fasciola spp. present in the<br>
western part of Iran. In the present study, 45 samples were collected from<br>
slaughterhouses in three provinces in the western part of Iran, including<br>
Ilam, Lorestan and Kermanshah.<br>
Methods: The flukes were detected using morphological methods and the<br>
DNA of all samples was extracted. The ribosomal internal transcribed<br>
spacer (ITS1) was identified by PCR and PCR-RFLP techniques using the<br>
Rsa1 restriction enzyme. Then, 15% of the samples were sequenced.<br>
Results: Based on their ITS1 sequence, all samples showed 700 bp bands.<br>
The results of sequencing showed a similarity of 99% to 100% across<br>
samples. Using the Rsa1 restriction enzyme, all samples produced three<br>
distinct bands (60 bp, 100 bp, and 360 bp). These results demonstrated<br>
that the most common fasciola in the western part of Iran in sheep and<br>
cattle is F. hepatica.<br>
Conclusion: The results of the present study showed that only the species<br>
F. hepatica is parasitizing livestock in the western part of Iran. Further<br>
studies using new molecular markers for more accurate identification of<br>
fasciola-causing species will be useful in the control and prevention of<br>
fascioliasis.<br>
